At the Central Park Zoo,
Marty the zebra (Chris Rock) is celebrating his tenth birthday, but longs to see the rest of the world from outside his
pampered life at the zoo, believing that he can find wide-open spaces to run around in, like Connecticut. Marty's best
friend, Alex the lion (Ben Stiller) attempts to cheer up his friend by singing Frank Sinatra's "New York, New York" with
him. Still unsatisfied, Marty gets some tips from the zoo's penguins; Skipper (Tom McGrath), Kowalski (Chris Miller),
Rico (John DiMaggio), and Private (Christopher Knights). The penguins are similarly trying to escape the zoo. Marty's
friends - Alex the lion, Melman the giraffe (David Schwimmer), and Gloria the hippopotamus (Jada Pinkett Smith) -
realize Marty's folly and try to follow him. The four, along with the penguins and the chimpanzees Mason (Conrad Vernon)
and his silent friend Phil eventually find themselves at Grand Central Station, but are quickly sedated by tranquilizer
darts when Alex's attempt to communicate with humans is mistaken for aggression. The zoo (under pressure from
animal-rights activists) is forced to ship the animals, by sea, to a Kenyan wildlife preserve. During their travels, the
penguins escape from their enclosure and take over the ship, intent on taking it to Antarctica. Their antics on the
bridge cause the crates containing Alex, Marty, Melmen, and Gloria to fall off the boat and wash ashore on
Madagascar.
The animals are soon able to regroup, initially believing themselves to be in San Diego, California.
Upon exploring, however, they come across a pack of lemurs led by King Julien XIII (Sacha Baron Cohen), and quickly
learn of their true location. Alex blames Marty for their predicament and attempts to signal for help to get back to
civilization. Marty, on the other hand, finds the wild to be exactly what he was looking for, with Gloria and Melman
soon joining him in enjoying the island. Alex eventually comes around, though his hunting instincts begin to show; he
has been away from the pampered zoo life of preprepared steaks for too long. The group is accepted by the lemurs, though
King Julien's adviser, Maurice (Cedric the Entertainer), cautions them about Alex's predatory nature. King Julien
ignores Maurice's concerns and persuades the group to help the lemurs fend off the fossa, who hunt the lemurs as prey.
While initially Alex scares the fossa away and is worshiped by the lemurs, he becomes delusional because of his
starvation and attacks Marty. Unable to overcome his instincts and hunger, King Julien banishes Alex to the far side of
the island, where the fossa live. Marty begins to regret seeing what Alex has turned into.
The penguins, having
been to Antarctica and found that it "sucks," land the boat on Madagascar. Seeing this as a chance to return Alex to New
York, Marty rushes after his friend against the wishes of the others. When Marty approaches the now grizzled, starving
Alex in his hut, Alex urges Marty to leave in fear of hurting his friend. Marty tries to get Alex to cooperate by
singing "New York, New York," but to no avail. The penguins, Gloria and Melman go to find Marty, but become trapped by
the fossa. At the last minute, Alex suddenly appears and approaches Marty as if he were prey, but then turns on the
fossa to scare them away from the lemur territory forever. Marty sees that Alex has overcome his predatory nature, and
the penguins help Alex satisfy his meat craving through sushi instead of steak. As the lemurs throw a bon voyage
celebration for the foursome, the penguins decide not to break the news that the ship has run out of fuel and that they
are still stuck on the island, setting the stage for Madagascar: Escape 2 Africa.

Un gruppo di pinguini, anche loro ospiti dello zoo, tentano di
scappare verso l'Antartide scavando una buca coi cucchiaini rubati, ma finiscono nel recinto di Marty, la zebra, che
comincia a pensare al mondo al di fuori delle mura dello zoo e vorrebbe scappare anche lui. Sembra che i suoi amici
siano riusciti a fargli cambiare idea, ma nella notte Marty fugge ed Alex, Gloria e Melman vanno a cercarlo.
Degli
animali selvaggi al centro di New York non passano inosservati, alla stazione centrale una vecchia picchia Alex e fa
scivolare Melman con la testa in un orologio e mentre Gloria cerca di liberarlo arrivano tutti i poliziotti, pompieri e
carabinieri che li isolano e Alex mentre cerca di parlagli credono che li stesse attaccando e così vengono catturati
e spediti in una riserva naturale in Kenya. Durante il viaggio i pinguini riescono a aprire il lucchetto con una
graffetta sputata da Rico,vanno in cabina e danno una pinnata al comandante e prendono il controllo della nave per
dirigersi verso l'Antartide, ma nella concitazione delle manovre le casse dei quattro personaggi si rovesciano in mare e
si ritroveranno poi nella spiaggia dell'isola di Madagascar. Qui vengono accolti da un gruppo di lemuri, capitanato da
Re Julien, un lemure istrionico ma simpatico, che tenta di usare i nuovi arrivati per difendersi dai fossa.
Ma nel
mezzo della "Natura" si riscoprono gli istinti animali e l'amicizia fra Marty ed Alex è messa a dura prova dalla fame
di quest'ultimo; più di una volta, infatti, il leone tenta di procurarsi il cibo mordendo la zebra. Alla fine quando
Marty si trova in una situazione di pericolo Alex riscopre il sentimento di amicizia che aveva verso di lui ed ad un
tratto i fossa inseguono Marty ma Melman piomba da una liana e lo salva e i fossa inseguono i 3, quando ad un tratto
Skipper con una pistola spara un fumo rosso e i fossa distratti guardano e loro con un timone li fanno fuori e Alex ad
un tratto arriva e tutto minaccioso e trasformato in un fossa fa credere a loro che Marty, Melman e Gloria sono sue
prede e fa fuggire i fossa via avendo quello che si meritano. Riusciranno poi a convivere quando scoprono che al leone
piace anche il pesce.
